人肝脂酶(HL)檢測試劑盒elisa技術流程
ELISA的基礎是抗原或抗體的固相化及抗原或抗體的酶標記.結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學活性,酶標記的抗原或抗體既保留其免疫學活性,又保留酶的活性.在測定時,受檢標本(測定其中的抗體或抗原)與固相載體表面的抗原或抗體起反應.用洗滌的方法使固相載體上形成的抗原抗體復合物與液體中的其他物質(zhì)分開.再加入酶標記的抗原或抗體,也通過反應而結(jié)合在固相載體上.此時固相上的酶量與標本中受檢物質(zhì)的量呈一定的比例.加入酶反應的底物后,底物被酶催化成為有色產(chǎn)物,產(chǎn)物的量與標本中受檢物質(zhì)的量直接相關,故可根據(jù)呈色的深淺進行定性或定量分析.由于酶的催化效率很高,間接地放大了免疫反應的結(jié)果,使測定方法達到很高的敏感度.ELISA可用于測定抗原,也可用于測定抗體.
然而,影響Elisa試驗結(jié)果的因素很多,故加強各個環(huán)節(jié)的質(zhì)量保證才能充分發(fā)揮其方法學的優(yōu)點.
試驗原理
試劑盒是固相夾心法酶聯(lián)免疫吸附實驗(ELISA).已知濃度的標準品、未知濃度的樣品加入微孔酶標板內(nèi)進行檢測.先將*標記的抗體同時溫育.洗滌后,加入親和素標記過的HRP.再經(jīng)過溫育和洗滌,去除未結(jié)合的酶結(jié)合物,然后加入底物A、B,和酶結(jié)合物同時作用.產(chǎn)生顏色.顏色的深淺和樣品中的濃度呈比例關系.
原理
免疫測定技術的基礎在于抗原抗體之間的特異結(jié)合反應,所以任何優(yōu)質(zhì)的診斷試劑離不開優(yōu)質(zhì)的原料,如抗原抗體,酶等.以前用于免疫測定的抗原通常為各種純化抗原,而抗體則為純化抗原免疫動物后獲得的多克隆抗體和使用雜交瘤技術得到的單克隆抗體,而抗原或抗體的標記物如酶標結(jié)合物則通過各種化學合成方法制備.近年來,隨著分子生物學的發(fā)展,使用基因工程方法制備各種特殊的抗原或抗體及其酶結(jié)合物等免疫測定試劑幾成為現(xiàn)實的新一代試劑,各種新型使用的測定方法也不斷出現(xiàn).

The Comprehensive Validation Support service includes all travel and labor charges for up to 20 days by the QIAGEN Validation Team. Typically, one week will be required to extract, quantify, and amplify the validation study samples and may require multiple individuals. Usually, two weeks is required to analyze the data and one week to write the final report. The expected time period to complete the Comprehensive Validation is six weeks. The actual time may vary depending on resource availability, including laboratory time dedicated to validation activities.
Lab Work – The QIAGEN specialist(s) will travel to the laboratory to conduct the on-site validation lab work. The QIAGEN specialist team will perform and complete all extractions, quantifications, amplfications, and capillary electrophoresis. The laboratory is responsible for providing systems/protocols that may be needed (generally laboratory SOPs, ex. interpretation guidelines). Additionally, the laboratory staff may be required to provide additional data needed for interpretation (ex. quantification/amplification/CE results) during the data review phase of the validation.
Once the final scope of the validation is determined, QIAGEN will provide a quote for all consumables required for the validation. Non-QIAGEN chemistry and accessories are not included and should be purchased to cover the validation experimental design including qPCR, STR, NIST standards, required samples types (blood, semen, buccal samples) and required substrates (swabs, materials, etc). The Customer is expected to have all materials on-hand 1 week prior to the validation start date.
Data Analysis – The Comprehensive Validation Support plan encompasses data collection (quants, amps, CE) during the on-site visit. The laboratory may be responsible for sharing (typically electronically via ) any validation data requested after the on-site visit. A QIAGEN specialist will complete all of the data analysis produced from the validation study design. This includes qPCR (quant) and STR (amp) analysis and linkage to starting samples.
Validation data CD/flashdrive – QIAGEN will review the validation data and draft a validation summary document, ensuring compliance with the FBI's QAS requirements/standards. QIAGEN will send validation data in an electronic format (/CD/flashdrive) to the laboratory. The laboratory is responsible for final compilation, any format change preferences, review and signoff of the validation report. The documentation provided by QIAGEN will aide in demonstrating compliance with all accreditation guidelines during laboratory audits. Should a hard copy of the validation study be preferred, the laboratory is required to specify prior to the onset of the validation.
Post-validation QIAGEN-to-Laboratory Transfer – Following completion of the validation and generation of the draft validation summary document, a designated member of the QIAGEN validation team will review the validation with the Lab Manager and the Technical Leader.
