DIY夾心法ELISA常見技術(shù)指南：
1、選擇抗體時選擇一個單抗和一個多抗進(jìn)行配對；若選擇兩個單抗，必須識別不同表位的抗體；從同一家公司選擇抗體對。
2、ELISA實驗中為了確定*信號和zui低背景應(yīng)將捕獲抗體（0.5-4μg/ml）和檢測抗體（0.25-2μg/ml）在預(yù)實驗中進(jìn)行彼此相抗的滴定。同時，應(yīng)包括在適當(dāng)范圍內(nèi)的標(biāo)準(zhǔn)品的系列稀釋。按試劑說明書常規(guī)提供的范圍進(jìn)行操作。
3、標(biāo)準(zhǔn)品的配制：對于每種細(xì)胞因子應(yīng)仔細(xì)閱讀說明書，注意每批的細(xì)節(jié)。在使用細(xì)胞因子前，瞬時離心試劑瓶，盡可能多地收回其中的細(xì)胞因子。根據(jù)每批說明書中的細(xì)節(jié)，將凍干的細(xì)胞因子復(fù)溶。
4、一般待測抗原標(biāo)準(zhǔn)曲線的線性范圍的可通過將標(biāo)準(zhǔn)品做8次2倍系列稀釋從2000pg/ml 到15pg/ml?？衫脴?biāo)準(zhǔn)的ELISA操作流程、放大試劑盒、第三類試劑、或改變酶底物系統(tǒng)可在一定范圍內(nèi)提高靈敏度。
5、為了優(yōu)化靈敏度，建議過夜孵育標(biāo)準(zhǔn)品和標(biāo)本。
6、若使用過氧化物酶作為顯色系統(tǒng)，應(yīng)嚴(yán)禁在洗液和稀釋液中加疊氮鈉，疊氮鈉可抑制過氧化物酶的活性。
7、當(dāng)測定混合液體中的抗原時，如血清，建議在標(biāo)本稀釋液中加不相干的Ig.
