重組增強(qiáng)型綠色熒光蛋白抗體保存條件:Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
重組增強(qiáng)型綠色熒光蛋白抗體,Anti-EGFP抗體溶解方法:
方法1:我們的抗體(凍干粉)已經(jīng)包含了0.01M PBS、1% BSA和0.1% 防腐劑(疊氮鈉或*),您只需要加入滅菌的雙蒸水就行了??贵w溶解后,置于-20℃保存,分裝后使用,避免反復(fù)凍融,可保證至少1年有效;
方法2:也可以只加入一半體積的雙蒸水,再用一半體積的甘油補(bǔ)足,置于-20℃保存,這樣抗體不會(huì)凍,有利于抗體的穩(wěn)定。
后續(xù)試驗(yàn)時(shí),再使用對(duì)應(yīng)的緩沖液稀釋成工作液,即用即配。
我們的抗體(凍干粉),在常溫放置下,至少一個(gè)月有效;若在-20℃下保存(*),至少三年有效。

免疫組化操作步驟:
(一)、儀器設(shè)備
1、 18cm不銹鋼高壓鍋或電爐或用微波爐
2、 水浴鍋
(二)、試 劑
1、 PBS緩沖液(pH7.2―7.4)
2、 0.01mol/L檸檬酸鈉緩沖液(CB,pH6.0,1000ml)
3、 3%甲醇―H2O2溶液:用30% H2O2 和80%甲醇溶液配制
4、 風(fēng)裱劑:
a. 甘油和0.5mmol/L碳酸鹽緩沖液(pH9.0–9.5)等量混合
b. 油和TBS(PBS)配制
5、 TBS/PBS pH9.0–9.5,適用于熒光纖維鏡標(biāo)本;pH7.0-7.4適合光學(xué)纖維標(biāo)本
(三)、操作流程
1、 脫蠟和水化:脫蠟前應(yīng)將組織芯片在室溫中放置60分鐘或60℃恒溫箱中烘烤20分鐘
a. 組織芯片置于二甲苯中浸泡10分鐘,更換二甲苯后在浸泡10分鐘
b. 無(wú)水乙醇中浸泡五分鐘
c. 95%乙醇中浸泡五分鐘
d. 75%乙醇中浸泡五分鐘
2、 抗原修復(fù):用于福爾馬林固定的石蠟包埋組織芯片

背景介紹:Enhanced green fluorescence protein (EGFP) is a 27 kDa protein derived from the jellyfish Aequorea victoria, which emits green light (emission peak at a wavelenth of 509 nm) when excited by blue light (excitation peak at a wavelenth of 395 nm). EGFP has become an invaluable tool in cell biology research, since its intrinsic fluorescence can be visualized in living cells. EGFP fluorescence is stable under fixation conditions and suitable for a variety of applications. EGFP has been widely used as a reporter for gene expression, enabling researchers to visualize and localize EGFP-tagged proteins within living cells without the need for chemical staining. Other applications of EGFP include assessment of protein protein interactions through the yeast two hybrid system and measurement of distance between proteins through fluorescence energy transfer (FRET) protocols. EGFP technnology has considerably contributed to a greater understanding of cellular physiology.
