產(chǎn)品介紹：The kinesins constitute a large family of microtubule-dependent motor proteins, which are responsible for the distribution of numerous organelles, vesicles and macromolecular complexes throughout the cell. Individual kinesin members play crucial roles in cell division, intracellular transport and membrane trafficking events including endocytosis and transcytosis. KIF1C is a member of the KIF1/Unc104 family of kinesin-like proteins, which are involved in the transport of mitochondria or synaptic vesicles in axons. The human KIF1A gene encodes a neuron-specific motor protein that delivers synaptic vesicle precursors to nerve terminals. KIF1A is a monomeric, globular molecule and has rapid anterograde motor activity (1.2 microns/s). KIF1A-mediated axonal transport plays a critical role in viability, maintenance and function of neurons, particularly mature neurons. KIF1A is associated with organelles that contain synaptic vesicle proteins such as synaptotagmin, synaptophysin and Rab 3A.

Anti-KIF1A抗體,微管驅(qū)動蛋白家族成員1A抗體保存條件：Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

1. 高熒光強(qiáng)度背景

（1）熒光標(biāo)記的抗體濃度應(yīng)該合適，如果濃度過高，背景會因為非特異性相互作用的增加而增加。

（2）在使用*抗體之前，將樣品與過量的蛋白一起培育，如小牛血清蛋白（BSA），脫脂干奶酪，或來自于同一寄主的正常血清作為標(biāo)記的第二抗體。這個步驟通過阻斷*抗體和細(xì)胞表面或胞內(nèi)結(jié)構(gòu)的非特異性的交互作用來降低背景。

（3）在使用*抗體之后，將樣品與5%至10%的來自于同一寄主的正常血清和作為標(biāo)記的第二抗體一起培育。這個步驟會減少不必要的第二抗體與*抗體、細(xì)胞表面或胞內(nèi)結(jié)構(gòu)之間的交互作用。

2. 細(xì)胞染色陽性但很弱

(1) 抗細(xì)胞因子抗體濃度不對或者熒光染料選擇不合適，增加抗體濃度，標(biāo)本表面抗原表達(dá)量低，應(yīng)該選擇熒光強(qiáng)度高的熒光染料，如PE，APC等。 (2)通透后細(xì)胞在胞內(nèi)染色前未洗, 按操作程序通透后洗細(xì)胞再胞內(nèi)染色。（3）抗體與固定后的胞內(nèi)抗原親和力低，需提高抗體濃度。

3、嚴(yán)重的細(xì)胞損失

（1）洗滌離心步驟丟失細(xì)胞，固定后細(xì)胞密度低于活細(xì)胞，需用高轉(zhuǎn)速離心，固定通透的細(xì)胞離心500g 。（2）加樣步驟丟失細(xì)胞或棄上清帶走細(xì)胞，應(yīng)小心吸樣。
