陰溝腸桿菌CMCC(B)45301子科現(xiàn)貨使用說(shuō)明書(shū):
1 復(fù)蘇菌種前應(yīng)準(zhǔn)備適于該菌種生長(zhǎng)的固體、液體培養(yǎng)基和培養(yǎng)設(shè)備，所有操作均應(yīng)在符合生物安全保護(hù)及無(wú)菌條件下進(jìn)行。
2 啟開(kāi)菌種陰溝腸桿菌 CMCC(B)45301前，用75%酒精棉消毒西林瓶表面。在無(wú)菌條件下，按鋁蓋上的箭頭方向打開(kāi)塑蓋，撕開(kāi)鋁蓋，打開(kāi)西林瓶膠塞，加入0.3mL左右的配套液體培養(yǎng)基，用無(wú)菌滴管反復(fù)吹吸，將凍干菌種溶解成為懸液，然后用無(wú)菌滴管或接種環(huán)移至斜面、平板或液體培養(yǎng)基，并連同剩余菌懸液的西林瓶一起放置于36℃培養(yǎng)箱中進(jìn)行培養(yǎng)。(菌種與培養(yǎng)和保存方法對(duì)應(yīng)表見(jiàn)附表1，培養(yǎng)基、培養(yǎng)方法和傳代保存方法見(jiàn)附表2)。
3 次日觀察菌種的生長(zhǎng)情況，如未生長(zhǎng)，應(yīng)繼續(xù)培養(yǎng)24h，或吸取培養(yǎng)之后的菌懸液中再次接種培養(yǎng)基，培養(yǎng)24h觀察。
4 菌株為一次性使用品，開(kāi)啟之后不能反復(fù)使用或保存，暫不開(kāi)啟的菌種應(yīng)在4℃保藏。
5 復(fù)蘇后的菌種在傳1-2代后使用，建議菌種使用5代后棄用。
注)：購(gòu)買(mǎi)后請(qǐng)盡量及時(shí)使用凍干菌種，如發(fā)現(xiàn)菌種不活或污染等情況，請(qǐng)?jiān)谫?gòu)買(mǎi)之日起1個(gè)月內(nèi)與我們。
